In the absence of arsenite, the ArsR repressor binds to its DNA binding site  and prevents expression of downstream genes. When arsenite enters the cell, binds the ArsR repressor leading to a conformational change which results in the dissociation of the ArsR protein from its binding site. Subsequently, the expression of downstream genes is promoted.
