Petunia hybrida T16 are transplastomic plants that express the aadA and the uidA gene. The LMO was constructed for an evaluation of pollen mediated transfer rates of transgenes located in the plastid genome in field grown transgenic petunia.

While the aadA gene is used as a selection marker (resistance towards streptomycine and spectinomycine is conferred) the expression of the uidA gene in plastids enables visualisation of transgenic plastids. Furthermore the homologous recombination of the insert into the plastidial genome was mediated by the rbcL- and the accD-gene so that exclusively plastids were transformed but not the nuclear genome. Since pollen does not possess plastids a vertical gene transfers seems impractical.

Notes regarding the genetic elements introduced or modified in this LMO
The accD and the rbcL genes encompass the insert and enable homologous recombination into plastidial DNA of Petunia hybrida Juss. T16.

The uidA gene is used for the colorimetric and fluorimetric detection of transformed plastids.
