This thermostable Beta-1,3-1,4-glucanase is the chimeric product
resulting from the recombination of two Beta-1,3-1,4-glucanase from
Bacillus amyloliquefaciens and Bacillus macerans.
The resulting protein depolymerises glucans in the aleurone layer
and the endosperm during caryopsis germination.
- Changes in quality and/or metabolite content
The gene introduced into barley was cloned by intragenic
recombination of two (1,3-1,4)-β-glucanases from Bacillus
amyloliquefaciens and Bacillus macerans. The enzyme
is composed of amino acids 1-12 of the glucanase from Bacillus
amyloliquefaciens and amino acids 13-214 of glucanase from
Bacillus macerans. Tyrosin at position 13 was deleted. The
nucleotide sequence was codon-optimized to ensure functionality of
the microbial gene in plants by raising the GC content to 63%.
