FLAVR SAVR™ tomato
Several lines
The FLAVR SAVR™ tomato lines were genetically engineered to express delayed softening by insertion of a PG encoding gene in the “antisense” orientation, which causes the endogenous PG messenger RNA (mRNA) to be degraded and, consequently, less production of PG protein.

The measured level of endogenous PG activity in transgenic FLAVR SAVR tomato was found to be less than 1% of PG activity found in the unmodified recipient organism.
The term “Recipient organism” refers to an organism (either already modified or non-modified) that was subjected to genetic modification, whereas “Parental organisms” refers to those that were involved in cross breeding or cell fusion.

Notes about the Other(s) sequence(s) specific to this LMO

The pg gene from tomato was introduced in the anti-sense orientation to suppress the expression of the endogenous pg gene.

Depending on the construct/vector used (see below), the antisense pg gene was under regulatory control of a single or two tandem copies of the 35S promoter from the cauliflower mosaic virus (CaMV). The terminator sequences for the antisense pg gene were the tml gene (T-tml) and the transcript 7 gene from the octopine-type Ti plasmid pTiA6 from A. tumefaciens.

Expression of the nptII gene was under the control of the 5' promoter and 3' terminator sequences from the mannopine synthase gene derived from A. tumefaciens or the 35s promoter and T-tml terminator, depending on the construct introduced in each line.

The DNA cassettes in two of the vectors used are shown below. Information on the cassettes in other vectors used to modify FLAVR SAVR™ was not found.

As a result of the introduction of a pg gene in the anti-sense orientation, the expression of the endogenous pg gene was suppressed.

The expression of other genes may have been affected in FLAVR SAVR™ tomato as a result of the transformation.
