The Chtra intron is derived from the endogenous transformer (tra) gene of Cochliomyia hominivorax, a master regulator of female sexual development in calliphorid flies. The tra gene is processed through sex-specific alternative splicing, producing distinct male and female transcripts. In males, inclusion of a male-specific exon (m1) containing multiple premature stop codons results in a non-functional transcript, whereas females generate a functional TRA protein through alternative splicing.

Because of these sex-specific splicing properties, the Chtra intron has been incorporated into transgenic genetic sexing systems developed for C. hominivorax. When positioned within or adjacent to a protein-coding sequence, the element causes sex-dependent transcript processing. In males, inclusion of the male-specific exon interrupts translation of the downstream coding sequence; in females, correct splicing permits production of the encoded protein. This feature enables female-specific lethality or sex separation strategies used in genetic control programs based on the Sterile Insect Technique (SIT).

The element has been used in genetically engineered strains of C. hominivorax to restrict expression of lethal effector genes to females, thereby allowing the production of male-only cohorts for release in population suppression programs. Gene constructs incorporating the Chtra splicing element have been described in transgenic sexing strains and female-lethal systems developed for New World screwworm control.

The genetic element is naturally occurring and is not itself synthetic or protein-coding. Its biological function is regulatory, acting through alternative RNA splicing mechanisms rather than encoding a protein product.
