Insect-resistant soybean
EN
COR-23134-4
Oui
COR-23134-4
The soybean (Glycine max) was modified to confer protection against lepidopteran pests and resistance to acetolactate synthase (ALS)-inhibiting herbicides. To achieve protection against susceptible lepidoptera, the LM soybean expresses the insecticidal proteins encoded by the Cry genes modified from Bacillus thuringiensis, cry1B.34.1 and cry1B.61.1, and the Adiantum trapeziforme insecticidal protein gene, ipd083Cb. The Cry proteins function as insecticidal toxins that, when ingested, are proteolytically activated in the midgut of the insects and bind to specific receptors on epithelial cells, forming pores that lead to insect death. The mode of action of the IPD083Cb protein is similar to that of Cry proteins, the IPD083Cb protein binds to specific sites in the midgut of susceptible insects, causing the insects to die. To achieve herbicide resistance, the LM soybean expresses the gm-hra_1 gene which is a variant of the acetolactate synthase enzyme derived from Glycine max (also known as acetohydroxy acid synthase). This protein contains two mutations in its amino acid sequence that reduce binding affinity to ALS-inhibiting herbicides and thus are responsible for GM-HRA resistance to ALS-inhibiting herbicides (e.g., sulfonylureas and triazolopyrimidines) and was used as a selection marker during the development of the transformation event. Four expression cassettes were incorporated into the modified soybean: three expressing the insecticidal proteins to confer both insect resistance, and one cassette expressing the GM-HRA protein to confer herbicide resistance and used as a marker selection.

Information of the inserted DNA sequences
The transforming plasmid PHP90315 contained a transfer DNA (T-DNA) region comprising four gene cassettes expressing the synthetic insecticidal proteins Cry1B.34.1 and Cry1B.61.1 modified from Bacillus thuringiensis, IPD083Cb derived from Adiantum trapeziforme, and the herbicide resistance protein variant of acetolactate synthase, GM-HRA_1 from Glycine max. Transformation of soybean line 93Y21 was achieved through Agrobacterium tumefaciens-mediated transformation of immature cotyledons.

(1) Transcription of the cry1B.34.1 coding sequence is directed by the histone H2B promoter together with the 5′ untranslated region and intron from the ubiquitin 1 gene from Zea mays. Transcription is terminated by the Oryza sativa ubiquitin terminator. Expression of this cassette results in production of the Cry1B.34.1 protein, a chimeric gene comprised of modified sequences of B. thuringensis-derived Cry genes (cry1B and cry1Ca1), to confer resistance against susceptible lepidopteran pests.

(2) Transcription of the cry1B.61.1 coding sequence is directed by the Glycine max chlorophyll a/b binding protein promoter and 5′ untranslated region. Transcription is terminated by the synthetic os-T28 bidirectional terminator region from a NAC domain-containing gene and the methylenetetrahydrofolate reductase gene from Oryza sativa. Expression of this cassette results in production of the Cry1B.61.1 protein, which is a a modified B. thuringiensis Cry1B class insecticidal protein that contributes to protection against susceptible lepidopteran pests.

(3) Transcription of the ipd083Cb coding sequence from A. trapeziforme is directed by the Phaseolus vulgaris ubiquitin 2 promoter, including the 5′ untranslated region and intron. Transcription is terminated by the synthetic os-T17 bidirectional terminator region from a 5-enolpyruvyl shikimate-3-phosphate synthase gene and the ribosomal protein S10 gene from Oryza sativa. Expression of this cassette results in production of the IPD083Cb protein, derived from Adiantum trapeziforme, which contributes to protection against susceptible lepidopteran pests.

(4) Transcription of the gm-hra_1coding sequence is directed by the Glycine max S-adenosyl-L-methionine synthetase promoter, including the 5′ untranslated region and intron. Transcription is terminated by the 3' untranslated region (3' UTR) of the native acetohydroxy acid synthase gene from Glycine max. Expression of this cassette results in production of the GM-HRA_1 protein, a modified soybean ALS enzyme that confers resistance to acetolactate synthase (ALS)-inhibiting herbicides.

Additional terminator regions were included in the T-DNA to reduce potential transcriptional interference between adjacent expression cassettes. These include the ubiquitin terminator and actin terminator from Sorghum bicolor between the first and second cassettes, and the Sorghum bicolor phosphoenolpyruvate carboxylase 1 terminator and Arabidopsis thaliana ubiquitin 3 terminator between the second and third cassettes. An additional terminator sequence from the Zea mays globulin-1 gene was positioned adjacent to the right T-DNA border and the terminator of the 19-kDA zein gene from Zea mays was inserted adjacent to the left T-DNA border.

Notes

  • The results from Southern-by-Sequencing (SbS) analysis and Sanger sequencing indicated that a single copy of the intended insertion, with the expected organization, was integrated into the genome of soybean COR23134, with the exception of three small deletions.
    • A 21-bp deletion was identified at position 21,740 in the pv-ubi2 promoter of the ipd083Cb cassette relative to the intended T-DNA insertion sequence.
    • Two additional deletions were identified at the T-DNA borders: 26 bp at the right border and 185 bp at the left border of the insertion
  • Border-region deletions are commonly observed during Agrobacterium-mediated plant transformation as a result of DNA repair processes. As the left and right border regions are not part of the expression cassettes, these deletions are considered unlikely to affect the function of the inserted genetic elements.
  • Alignment of the SbS reads from soybean COR23134 and the non-GM control soybean line 93Y21 to the PHP90315 plasmid sequence confirmed the absence of integrated plasmid backbone DNA in soybean COR23134, including sequences encoding antibiotic resistance genes.
  • Bioinformatic analysis of the insertion site and flanking genomic DNA indicated that the insertion did not disrupt any known endogenous soybean genes.
