Modena potato with altered starch
EN
AV43-6-G7
Yes
AVE-436G7-1
Potato was modified by the insertion of a fragment of the granule-bound starch synthase gene (gbss) in the anti-sense direction to down-regulate the endogenous gbss gene.

The anti-sense repeat fragment of the gbss gene leads to a reduction of the amylose fraction in the starch of the potato tuber resulting in a concomitant increase in the levels of amylopectine.

The potato event AVE436G7-1 does not contain any selective genes for resistance to antibiotics or herbicides.

The term “Recipient organism” refers to an organism (either already modified or non-modified) that was subjected to genetic modification, whereas “Parental organisms” refers to those that were involved in cross breeding or cell fusion.

Some of these genetic elements may be present as fragments or truncated forms. Please see notes below, where applicable.
  • BCH-GENE-SCBD-14997-6 Granule bound starch synthase gene promoter | Solanum tuberosum (Potato, SOLTU)
    Promoter
  • BCH-GENE-SCBD-48072-3 Granule-bound starch synthase gene | Solanum tuberosum (Potato, SOLTU)
Protein coding sequence | altered carbohydrate composition: increased amylopectin content
  • BCH-GENE-SCBD-45875-7 Beta-galactosidase gene | Escherichia coli (ECOLX)
Protein coding sequence | Selectable marker genes and reporter genes
  • BCH-GENE-SCBD-100269-8 Nopaline Synthase Gene Terminator | Agrobacterium tumefaciens (Agrobacterium)
    Terminator
A truncated portion of the coding sequence of gbss gene (CS-gbss) was cloned in anti-sense orientation under control of the gbss gene promoter (P-gbss).

AVE-436G7-1 contains one insert of an inverted repeat construct of the potato gbss gene. This LMO also contains part of the beta-galactosidase gene (lac) gene from Escherichia coli. No elements outside of the left-border (LB) and right-border (RB) of the T-DNA were inserted. Both LB and RB sequences of the T-DNA were truncated during the insertion.

The portions of the GBSS gene sequence in the construct organization in this LMO is as follows:
- GBSS cDNA fragment; 5’ 1.1 kb part, Solanum tuberosum, sense orientation in relation to the promoter sequence.
- GBSS cDNA fragment; 3’ 1.3 kb part, Solanum tuberosum, antisense orientation in relation to the promoter sequence, functioning as spacer element.
- GBSS cDNA fragment; 5’ 1.1 kb part, Solanum tuberosum, antisense orientation in relation to the promoter sequence.

Detection method(s)
Southern blot showed one insertion site with one copy of the constructed was inserted into AVE-436G7-1. The insertion site in the recipient organism (potato variety "Karnico") in which the T-DNA was integrated in AVE-436G7-1 was found to have a deletion of 1814 bp.
