The siRNA target sequence was taken from a portion of EU974548 with multiple binding sites of the most abundant siRNAs present in tassel tissues. Deep sequencing of small RNA populations in tassels at different development stages elucidated the male specific siRNA populations.

The sequence shares homology to endogenous small interfering RNAs (siRNAs) abundant in male tissues (tassels). Thus, when siRNAs are produced in the male tissues, the RNA interference machinery will target the target sequence and cause degradation of the full transcript. This allows for an inducible, male tissue-specific system to be created, where expression will be silenced in the male tissues.
