Additional Information
PpoI is an intron-encoded endonuclease that specifically cleaves (*) double stranded DNA at the following recognition site:

5'- CTCTC TTAA*GGTAGC-3'
3'-GAGAG*AATT CCATCG-5'

The endonuclease creates staggered cuts with 5'-TTAA-3' overhangs. The enzyme itself functions as a homodimer and each submit has a molecular weight of 20 kDa (40 kDa as a homodimer). The target sequence can be found in ribosomal DNA repeats on the X chromosomes of certain insects and chromosome 12 of Saccharomyces cerevisiae. In biotechnology, this specific cutting on the X chromosome of insects could be utilized to promote a male sex bias (sex distorter drive) or male sterility.
