The sugarcane (Saccharum sp.) was modified with the commercial vector pCTC302 to obtain event CTC79005-2. The modified sugarcane contains expression cassettes for Bacillus thuringiensis serovar kustaki strain HD73 cry1Ac, which encodes a toxin of 615 amino acids (68 kDa - estimated molecular weight) and Escherichia coli nptII gene, which encodes the enzyme for the neomycin phosphotransferase II of 265 amino acids (29.2 kDa - estimated molecular weight). The expression of Cry1Ac in sugarcane confers resistance to Lepidoptera, such as cane borer (Diatraea saccharalis). The expression of NPTII confers resistance to kanamycin and geneticin, the later of which was used as a selectable marker during transformation.
The term “Recipient organism” refers to an organism (either already modified or non-modified) that was subjected to genetic modification, whereas “Parental organisms” refers to those that were involved in cross breeding or cell fusion.

The sugarcane contains two expression cassettes: Bacillus thuringiensis serovar kustaki strain HD73 cry1Ac and Escherichia coli neomycin phosphotransferase II (nptII). Both cry1Ac and nptII are under the control of a Zea mays ubiquitin1 promoter and an Agrobacterium tumefaciens nopaline synthase terminator. The promoter sequence additionally includes the first intron from the maize ubiquitin gene for enhanced gene expression. Due to the constitutive nature of the promoter, high levels of transcription are expected in all plant tissues.

- Cry1Ac is a toxin of 615 amino acids (68 KDa - estimated molecular weight). The cry1Ac gene present in the pCTC302 construct corresponds to a synthetic and truncated DNA sequence. The cry1Ac gene had its nucleotides synthetically optimized in order to use sugarcane preferred codons (also see Weng et al. 2011).
- NPTII is 265 amino acids in length (29.2 KDa - estimated molecular weight). The coding sequence was originally sourced from E. coli Tn5 transposon.
- The pCTC001 vector was used as a basis for cloning and multiplying the T-DNA containing the gene cassettes for this event. This base plasmid is a binary plasmid vector for use in E. coli and A. tumefaciens with an original total size of 6,429 basepairs. It contains the aadA gene, which confers resistance to the antibiotic spectinomycin; origin of replication for E. coli from plasmid pBR322, origin of replication for A. tumefaciens from plasmid pVS1, necessary transcriptional control elements, right and left borders for T-DNA transfer (nopaline type) and a multiple cloning site.
