The DGT-28 EPSPS from Streptomyces sviceus is part of a new class (class IV) of 5-enolpyruvylshikimate-3-phosphate synthase enzymes (EPSPS) that confer tolerance to high concentrations of glyphosate.
The enzyme is part of the shikimate pathway, which is involved in the biosynthesis of aromatic amino acids and other aromatic compounds. The anzyme catalyzes the transfer of the enolpyruvoyl moiety of phosphoenolpyruvate to the 5′-hydroxyl group of shikimate-3-phosphate with a concomitant liberation of inorganic phosphate. In susceptible plants, the herbicide glyphosate inhibits the biochemical activity of the enzyme.

Other classes of EPSPS enzymes:
- Class I EPSP synthases are typically sensitive to glyphosate and have been primarily characterized from plants and bacteria.
- Class II enzymes are found in microbes and possess varying natural levels of tolerance to glyphosate without compromising the binding of the native co-substrate phosphoenolpyruvate.
- Class III enzymes are also found in microbes but have less than or equal to 25% protein sequence identity compared to E. coli EPSPS and less than or equal to 22% identity to Agrobacterium sp. strain CP4 while exhibiting increased tolerance to high levels of glyphosate.
