Biotech Vac Salmonella is an oral recombinant veterinary vaccine intended to induce protective immunity against Salmonella caused by different species, including Salmonella enteritidis and Salmonella infantis. To achieve protection against Salmonella serotypes, B. subtilis expresses the TolB protein subunit which acts as the vaccine antigen and induces an immune response that contributes to protection the disease. The final vaccine contains no viable modified bacteria, as the recombinant B. subtilis is inactivated before formulation.
The term “Recipient organism” refers to an organism (either already modified or non-modified) that was subjected to genetic modification, whereas “Parental organisms” refers to those that were involved in cross breeding or cell fusion.

Information on the inserted DNA sequence
The modified Bacillus subtilis contains a recombinant pHT01 plasmid carrying a genetic expression cassette for the expression of the TolB antigen from Salmonella enterica subsp. enterica serovar Typhimurium strain LT2 used in the vaccine formulation. The DNA sequence encoding the Salmonella TolB protein was cloned into the pHT01 expression plasmid. The coding sequence was inserted into the BamHI/XbaI multiple cloning site of the expression plasmid.
Expression of the TolB coding sequence is regulated by a LacI/PgroESL regulatory cassette, in which the groESL operon is composed of the groEL chaperonin and groES helper protein from Bacillus subtilis and transcription is controlled by the LacI repressor encoded by the lacI gene and induced by isopropyl β-D-1-thiogalactopyranoside (IPTG). Production of the TolB protein is inactivated by treatment with formalin before being used as a vaccine. The final product is intended to be orally administered to poultry.

Note
- In order to confirm the construct, the recombinant pHT01 plasmid containing the cloned Salmonella TolB protein gene was first inserted into Escherichia coli TOPO 1 by direct ligation. After confirmation, it was inserted then into Bacillus subtilis 1A6 (ATCC® 39085TM) with the same method.
- The vector pHT01 is made up of 7,956 bp where the DNA sequence encoding the Salmonella TolB protein (tolB gene) is composed of 1,293 bp.
- Polymerase chain reaction, with primers that amplify a 1,346 bp fragment, is used to detect the presence of the recombinant plasmid in Escherichia coli and B. subtilis.
- For selection of positive transformants in Escherichia coli and B. subtilis, pHT01 contains selection markers for antibiotics resistance genes, ampicillin and chloramphenicol.
