The cotton (Gossypium hirsutum) MON15947 is a segregant derived from the cotton MON15985 (Bollgard II™) through conventional breeding, in which the MON531 (Bollgard™) cassette containing the Cry1Ac protein and the NPTII selection marker were segregated out. The MON15947 expresses the Cry2Ab2 crystalline protein from Bacillus thuringiensis subsp. kurstaki, which confers resistance to lepidopteran cotton pests, including the tobacco budworm (Heliothis virescens), pink bollworm (Pectinophora gossypiella), cotton bollworm (Helicoverpa zea), and soybean looper (Pseudoplusia includens).
The term “Recipient organism” refers to an organism (either already modified or non-modified) that was subjected to genetic modification, whereas “Parental organisms” refers to those that were involved in cross breeding or cell fusion.

Information on the inserted DNA sequences
The cry2Ab2 coding sequence is under the control of an enhanced cauliflower mosaic virus (CaMV) 35S promoter and the nopaline synthase (nos) terminator from Agrobacterium tumefaciens. The expression cassette also includes a 5′ leader sequence derived from the HSP70 gene of Petunia hybrida, intended to enhance protein expression, and a chloroplast transit peptide (TS-CTP2) from Arabidopsis thaliana, which directs the translated protein to the chloroplast. The uidA coding sequence is under the control of the same enhanced 35S promoter and nos terminator.

Like other delta-endotoxins derived from Bacillus thuringiensis, the Cry2Ab2 protein exerts its insecticidal activity by binding to specific receptors located in the midgut epithelium of susceptible lepidopteran insects. Upon binding, specific cation pores form, disrupting the intestinal ionic balance, causing intestinal paralysis and eventual insect death. The Cry2Ab2 protein is highly selective and is only active against lepidopteran pests of cotton, including the tobacco borer (Heliothis virescens), pink boll (Pectinophora gossypiella), cotton bollworm (Helicoverpa zea), and soybean looper (Pseudoplusia includens).
In addition to the Cry2Ab2 protein that confers insect resistance, the MON-15947-5 event contains the Escherichia coli Beta-glucuronidase (GUS/uidA) visual marker gene, which allows the identification of transformed plants by means of a colorimetric assay that produces a blue color in the transformed cells.

The inserted cassette in MON15947 corresponds to the insertion previously obtained by transformation with the KpnI fragment of the PV-GHBK11 vector. The insert contains 5,719 bp derived from this fragment. Relative to the original KpnI fragment, 307 bp at the 5′ end and 66 bp at the 3′ end are absent in the transformed plant. The deduced amino acid sequence of the cry2Ab2 coding region is as expected based on the PV-GHBK11 sequence; however, the inserted β-D-glucuronidase sequence differs by a single amino acid substitution (E377K).
