Potato with altered starch composition
EN
AM99-2003
No
Fragments of the genes of the starch branching enzymes, be1 and be2, were introduced to modify the starch composition in potato tubers, thus raising the level of amylose.

EN
The term “Recipient organism” refers to an organism (either already modified or non-modified) that was subjected to genetic modification, whereas “Parental organisms” refers to those that were involved in cross breeding or cell fusion.
Cultivar/breeding line: Dinamo

Characteristics of the modification process
Derived from pGTV-KAN
EN
  • Agrobacterium-mediated DNA transfer
Some of these genetic elements may be present as fragments or truncated forms. Please see notes below, where applicable.
  • BCH-GENE-SCBD-14997-6 Granule bound starch synthase gene promoter | Solanum tuberosum (Potato, SOLTU)
    Promoter
  • BCH-GENE-SCBD-48366-2 Branching Enzyme 1 | Solanum tuberosum (Potato, SOLTU)
Protein coding sequence | altered carbohydrate composition: increased amylopectin content
  • BCH-GENE-SCBD-48453-2 Branching Enzyme 2 | Solanum tuberosum (Potato, SOLTU)
Protein coding sequence | altered carbohydrate composition: increased amylopectin content
  • BCH-GENE-SCBD-100269-8 Nopaline Synthase Gene Terminator | Agrobacterium tumefaciens (Agrobacterium)
    Terminator
  • BCH-GENE-SCBD-100270-6 Nopaline Synthase Gene Promoter | Agrobacterium tumefaciens (Agrobacterium)
    Promoter
  • BCH-GENE-SCBD-15001-5 Neomycin Phosphotransferase II | Escherichia coli (ECOLX)
Protein coding sequence | Resistance to antibiotics (Kanamycin)
  • BCH-GENE-SCBD-103067-9 Transcript 7 gene 3' untranslated region | Agrobacterium tumefaciens (Agrobacterium)
    Terminator
In potato line AM99-2003 a gene consisting of gbss promoter from Solanum tuberosum, the coding region fragments of be1 and be2 (starch-branching enzyme) in tandem and antisense orientation relative to the promoter and the nos polyadenylation sequence from Agrobacterium tumefaciens have been inserted into potato variety Dinamo thus reducing the amount of amylopectin in the starch fraction of the tuber.

In addition the neomycin phosphotransferase gene (nptII) connected to the Agrobacterium tumefaciens nopaline synthase promoter and g7 polyadenylation sequence from Agrobacterium tumefaciens has been inserted as selectable marker gene conferring resistance to kanamycin.
