Soybean (Glycine max) was modified to confer tolerance to multiple herbicides. For resistance to 2-methoxy-3,6-dichlorobenzoic acid (dicamba), the soybean expresses Stenotrophomonas maltophilia dicamba monooxygenase, which degrades the herbicidal compound through oxygenation. For tolerance to glufosinate, the soybean expresses Streptomyces viridochromogenes phosphinothricin N-acetyltransferase, which inactivates the herbicidal compound through acetylation. For tolerance to quizalofop and 2,4-dichlorophenoxyacetic acid herbicides, the soybean expresses Sphingobium herbicidovorans R-2,4-dichlorophenoxypropionate dioxygenase, which inactivates the herbicides through oxygenation. The enzyme may also act on other aryloxyphenoxypropionate and synthetic auxin herbicides as well. For tolerance to β-triketone herbicides (e.g., mesotrione and benzobicyclon), the soybean expresses Oryza sativa triketone dioxygenase, which catalyzes hydroxylation of the herbicidal compounds to inactivate them. The modified soybean does not contain any selectable markers (splA and aadA expression cassettes) as they were crossed out of the line during development of the commercial line.

The modified soybean contains four gene cassettes: Stenotrophomonas maltophilia dicamba monooxygenase (dmo), Streptomyces viridochromogenes phosphinothricin N-acetyltransferase gene (pat), Sphingobium herbicidovorans R-2,4-dichlorophenoxypropionate dioxygenase (ft_t.1) and Oryza sativa triketone dioxygenase (tdo).
Transcription of dmo is under the control of an Arabidopsis thaliana ubiquitin 3 promoter and a Medicago truncatula sali3-2 protein terminator. An A. thaliana albino and pale green 6 target peptide was included to direct the translated protein to the chloroplast. Due to the constitutive nature of the promoter, high levels of transcription are expected.
Transcription of pat is under control of a synthetic promoter GSP579 and a M. truncatula heat shock protein 20 terminator. A synthetic intron (GI102) was included to enhance expression of the pat sequence. Due to the constitutive nature of the promoter, high levels of transcription are expected.
Transcription of ft_t.1 is under control of an A. thaliana ubiquitin 10 promoter and a M. truncatula gene of unknown function 1 terminator. Due to the constitutive nature of the promoter, high levels of transcription are expected.
Transcription of tdo is under control of a synthetic promoter GSP579, a synthetic intron GSI102 and a synthetic terminator GST7. Given the constitutive nature of promoter and enhancement by the other synthetic regulatory elements, high levels of expression are expected.

Note:
- The ubiquitin 10 promoter contains leader and intron sequences.
- The tdo sequence was codon-optimized for expression in soybean.
- Next-generation sequence analyses indicated that the line contains a single T-DNA insert without integration of vector backbone sequences. Directed sequence analyses indicated that the genetic cassettes were presented as expected (without rearrangement).
- A second T-DNA was inserted during the production of MON94313. The second T-DNA contained selectable marker cassettes for 3'(9)-O-nucleotidyltransferase (aadA) and sucrose phosphorylase (splA). Subsequent segregation, selection and screening were used to isolate those plants that did not contain the second T-DNA insertion following the crossing of T1 generation progeny. See attached regulatory applications for more details.
