Blue-purple moth orchid
The moth orchid (Phalaenopsis sp.) has been modified for altered floral coloration. To achieve the blue-purple flowers, the orchid expresses Commelina communis flavonoid 3',5'-hydroxylase, which promotes the synthesis of anthocyanin pigments and results in changed petal coloration. In addition, the orchid expresses Escherichia coli hygromycin B phosphotransferase for hygromycin selection during transformation.
The term “Recipient organism” refers to an organism (either already modified or non-modified) that was subjected to genetic modification, whereas “Parental organisms” refers to those that were involved in cross breeding or cell fusion.
Characteristics of the modification process
  • Agrobacterium-mediated DNA transfer
Some of these genetic elements may be present as fragments or truncated forms. Please see notes below, where applicable.
  • BCH-GENE-SCBD-101416-6 Ti plasmid right border repeat | Agrobacterium tumefaciens (Agrobacterium)
    Plasmid vector
  • BCH-GENE-SCBD-100287-7 CaMV 35S promoter | Cauliflower mosaic virus (CaMV)
  • BCH-GENE-SCBD-14991-8 Hygromycin B phosphotransferase gene | Escherichia coli (ECOLX)
Protein coding sequence | Resistance to antibiotics (Hygromycin),Selectable marker genes and reporter genes
  • BCH-GENE-SCBD-100269-8 Nopaline Synthase Gene Terminator | Agrobacterium tumefaciens (Agrobacterium)
  • BCH-GENE-SCBD-104820-3 Omega 5' untranslated leader | Tobacco mosaic virus (TMV)
  • BCH-GENE-SCBD-100290-6 CaMV 35S terminator | Cauliflower mosaic virus (CaMV)
  • BCH-GENE-SCBD-101415-9 Ti plasmid left border repeat | Agrobacterium tumefaciens (Agrobacterium)
    Plasmid vector
  • BCH-GENE-SCBD-258648-1 Flavonoid 3',5'-hydroxylase | Commelina communis (Asiatic dayflower, Dayflower, Common dayflower, Asiatic daisy)
Protein coding sequence | Changes in quality and/or metabolite content (Pigmentation / Coloration)
The modified orchid contains two gene cassettes: Escherichia coli hygromycin B phosphotransferase (hpt) and Commelina communis flavonoid 3', 5'-hydroxylase (f3'5'h).

The hpt coding sequence is under control of the Cauliflower mosaic virus (CaMV) 35S promoter and Agrobacterium tumefaciens nopaline synthase terminator. Due to the constitutive nature of the promoter, high levels of transcription and gene expression of hpt are expected.

The f3'5'h coding sequence is under control of a CaMV 35S promoter and terminator. A 5' untranslated region of Tobacco mosaic virus is also present between the CaMV promoter and the f3'5'h coding sequence. This sequence improves the translation of the F3'5'H. Due to the constitutive nature of the promoter, high levels of transcription and gene expression of f3'5'h are expected.
