The moth orchid (Phalaenopsis sp.) has been modified for altered floral coloration. To achieve the blue-purple flowers, the orchid expresses Commelina communis flavonoid 3',5'-hydroxylase, which promotes the synthesis of anthocyanin pigments and results in changed petal coloration. In addition, the orchid expresses Escherichia coli hygromycin B phosphotransferase for hygromycin selection during transformation.

The modified orchid contains two gene cassettes: Escherichia coli hygromycin B phosphotransferase (hpt) and Commelina communis flavonoid 3', 5'-hydroxylase (f3'5'h).

The hpt coding sequence is under control of the Cauliflower mosaic virus (CaMV) 35S promoter and Agrobacterium tumefaciens nopaline synthase terminator. Due to the constitutive nature of the promoter, high levels of transcription and gene expression of hpt are expected.

The f3'5'h coding sequence is under control of a CaMV 35S promoter and terminator. A 5' untranslated region of Tobacco mosaic virus is also present between the CaMV promoter and the f3'5'h coding sequence. This sequence improves the translation of the F3'5'H. Due to the constitutive nature of the promoter, high levels of transcription and gene expression of f3'5'h are expected.
