Aminolevulinic acid synthase

Aminolevulinic acid synthase is the rate-limiting enzyme for heme biosynthesis. Because oxidized forms of downstream tetrapyrrole intermediates (uroporphyrin III, coproporphyrin III, and protoporphyrin IX),exhibit cytotoxic photodynamic properties, cellular levels of this enzyme are tightly regulated. Consequently, aminolevulinic acid synthase is maintained at low abundance under normal conditions; however, when intracellular heme levels are reduced, its expression increases several-fold to restore heme homeostasis. The regulation of aminolevulinic acid synthase by heme thus represents a classical example of end-product feedback control.
