Rubber tree modified to express a reporter gene

Hevea brasiliensis was modified to express the glucuronidase (GUS) reporter gene encoding glucuronidase enzyme that hydrolyzes x-gluc - a colourless substrate, into soluble blue coloured product. This serves as an indicator of successful expression of transgene product in the target tissue.

GUS enzyme activity has been detected in latex and leaf tissue of transformed Hevea plants.

Hevea transformed with GUS also harbours the nptII gene that confers resistance to kanamycin, which was incorporated in the growth media to select transformed callus after co-cultivation with Agrobacterium.

Agrobacterium tumefaciens containing p35S-GUS-INT plant expression vector was a gift from Prof. L. Willmitzer, Institut fur Genbiologische Forschung, Berlin.

The inserted genetic material is the region between the left border and right boarder (T-DNA) from the Ti plasmid of Agrobacterium tumefaciens harbouring the above described gene construct.
