The modified hybrid poplars (Populus canescens; Populus tremula x Populus alba) have an altered wood composition resulting from the introduction of a knock-out mutation in the endogenous transaldolase genes tra2a and tra2b. These genes are involved in the pentose phosphate pathway that is upstream of the monolignol pathway, which is the basis for the biosynthesis of lignin. The modified trees additionally contain higher levels of cellulose in their cell walls and overall wood composition. The trees also contain a non-functional CRISPR-associated endonuclease from Streptococcus pyogenes and an Escherichia coli hygromycin B phosphotransferase selectable marker.

The term “Recipient organism” refers to an organism (either already modified or non-modified) that was subjected to genetic modification, whereas “Parental organisms” refers to those that were involved in cross breeding or cell fusion.

  • Agrobacterium-mediated DNA transfer
  • Gene editing (e.g. CRISPR-Cas, etc.)
Some of these genetic elements may be present as fragments or truncated forms. Please see notes below, where applicable.
The modified poplars contain cassettes for Streptococcus pyogenes CRISPR-associated endonuclease 9 (cas9), synthetic guide RNAs and Escherichia coli hygromycin B phosphotransferase (hph).

The cas9 coding sequence is under control of a Cauliflower mosaic virus 35S promoter and a Pisum sativum ribulose-1,5-bisphosphate carboxylase/oxygenase small subunit terminator. The cas9 sequence contains a mutation to make the final protein product non-functional. During the transformation of the poplars, the CAS9 protein was initially active. However, with the presence of the guide RNA targeting its own sequence, a mutation was induced to produce a final non-functional protein product.

The hph coding sequence is under control of the Agrobacterium tumefaciens nopaline synthase promoter and terminator.

Note:
  • Information was not provided about the promoters for the guide RNA sequences.
  • No vector backbone sequences were integrated into the poplar genome.

LMO characteristics
After transformation, the functional CAS9 protein complexes with the guide RNAs to induce mutations in the endogenous transaldolase 2a and transaldolase 2b, as well as the cas9 sequence itself. The genome editing results in gene products that are not functional.
