Maize 3272 was developed through genetic transformation mediated by Agrobacterium tumefaciens, by inserting the synthetic gene amy797E, whose expression produces the thermostable alpha-amylase protein AMY797E. The amy797E gene is a chimeric gene derived from sequences of three alpha-amylase genes originating from three hyperthermophilic microorganisms of the archaea order Thermococcales. This alpha-amylase chimeric gene was assembled from parental sequences using GeneReassembly™ technology (Diversa Corporation, San Diego, CA) and was selected for the thermostability properties of the encoded amylase protein required during the liquefaction phase of the starch in the processing of corn grains, such as in the production of ethanol. Maize 3272 also contains the pmi gene from E. coli that encodes a phosphomannose isomerase and is under regulation by the promoter of the ubiquitin gene from Zea mays and the NOS terminator from Agrobacterium tumefaciens. This gene acts as a selection marker in the transformation process.

Methodology and points to consider
Corn 3272 has a history of safe use and is approved for human and animal consumption by CTNBio in Brazil. Corn 3272 does not pose significant risks to human, animal or environmental health.

The proteins introduced in Maize 3272 come from organisms with a consolidated and safe history of use; being well studied regarding the mechanisms of action and absence of anti-nutritional, toxic, allergenic activities, which may alter performance or cause any type of damage to any species that consume them. The nutritional composition of this corn is equivalent to the composition of corn varieties that have not been genetically transformed, except for the expression of AMY797E and PM proteins.

Corn 3272 has been approved for use in human food in 18 countries, of which 13 have also approved for use in animal feed. The cultivation of Maize 3272, object of this application, was approved by three countries: Canada (2008), Japan (2010) and the United States (2008), with no adverse effects reported.

Corn 3272 is a well-known and studied crop, and does not have any of the attributes usually associated with weeds, with a wide history of safe use and consumption; b) Corn 3272 presents the same chemical composition of analyzed analytes, based on the international literature, when compared to conventional commercially cultivated corn hybrids, not affecting the degradability of Corn 3272 plants compared to non-genetically modified corn; c) Corn 3272 has the same agronomic characteristics as conventional non-genetically modified corn, thus, no changes are expected in the dispersal capacity of propagation and reproduction structures, survival capacity and long-term reproduction structures; nor is it expected to change its ability to add to or remove substances from the soil; d) when it is cultivated, Maize 3272 does not cause a different impact on non-target organisms, compared to the cultivation of a non-genetically modified corn.

Corn 3272 are substantially equivalent in characteristics agronomic characteristics to the corresponding quasi-isogenic non-transgenic hybrids, in addition to the characteristics introduced, grain yield and other agronomic measurements were compared in trials at various locations. The agronomic evaluation tests were carried out in the field in 4 locations in Brazil, in the 2020/21, in the municipalities of Bandeirantes - SP, Palmeiras de Goiás - GO, Juazeiro - BA, and Mandirituba - PR.

CIBio from Syngenta Seeds Ltd. considered that the cultivation, commercialization or use in human and animal food are not activities that are potentially causing significant degradation of the environment or cause harm to human and animal health.

CTNBio considered that the commercial release for planting genetically modified corn 3272 complies with the relevant norms and legislation that aim to guarantee the biosecurity of the environment, agriculture, human and animal health. Thus, meeting the conditions described in the process and in this technical opinion, this activity is not potentially causing significant degradation of the environment or damage to human or animal health.
