Late blight-resistant Jalene potato

The potato (Solanum tuberosum) was modified to express three late blight resistance genes. To achieve this, the potato expresses Solanum bulbocastanum Phytophthora infestans resistance gene blb1, S. bulbocastanum Phytophthora infestans resistance gene blb2 and Solanum venturii Phytophthora infestans resistance gene 1 (Rpi-vnt1.1), which recognize pathogen proteins to mediate a hypersensitive defense response. In addition to the resistance genes, the potato expresses Escherichia coli neomycin phosphotransferase, which was used for kanamycin selection during transformation.

Note: For the transformation event, a line has not be selected. Thus, a generic transformation event was chosen.

The modified potato (Solanum tuberosum) contains four gene cassettes: Escherichia coli neomycin phosphotransferase (nptII); Solanum venturii Phytophthora infestans resistance gene 1 (Rpi-vnt1.1); Solanum bulbocastanum Phytophthora infestans resistance gene blb1 (Rpi-blb1; RB) and S. bulbocastanum Phytophthora infestans resistance gene blb2 (Rpi-blb2)

The nptII coding sequence is under control of a Cauliflower mosaic virus 35S promoter and terminator.

The Rpi-blb1, Rpi-blb2 and Rpi-vnt1.1 coding sequences are under control of their native promoters and terminators.

Note:
  • The genetic element sizes of the promoters and terminators of Rpi-blb1, Rpi-blb2 and Rpi-vnt1.1 are not clear.
