The genetically modified potato plants constitutively express a mutated fragment of the NIb gene of the Potato Virus Y, which, in its native form, codes for a RNA-dependent RNA polymerase. The modification is expected to confer resistance to PVY.

To enable the detection the Nib gene product, the modified NIb gene was fused to the enhanced blue fluorescent protein gene from the jellyfish Aequorea victoria as a reporter gene.

A mutated sequence of the NIb replicase of the Potato Virus Y was fused to the enhanced blue fluorescent protein gene. They are expressed under the control of the 35S promoter and termination sequence of the Cauliflower Mosaic Virus.

The mutated sequence has a deletion at the C-terminus (approx. 400 nt) and at the N-terminus (6 nt), and an internal shift in the reading frame affecting 14 amino acids, leading to a non-functional gene product.
